手自一體變速箱的檔位有P,R,N,D和M/S。P,R,N和D檔和純自動檔的沒有什么區別,不同點是M/S檔,這個檔是手動模式檔(有的也叫運動模式)。這里主要提一提S擋。掛進S擋后電腦的換擋邏輯會產生變化，相對來說升擋會比較遲，但動力更強。

使用S擋時，把檔桿向+的方向推動是進檔,向-的方向是退檔，不需要踩剎車，有些車的手動模式會設置5000轉強制進檔。雖然S擋動力更強，但并不建議車主長時間使用該檔位。因為該檔位雖然能夠瞬間提高轉速加大動力，但長期使用卻會讓自動變速箱承受更多的負荷，導致變速箱過早磨損。
